Crystal Structure of Soybean Lipoxygenase L-1 at 1.4 Å Resolution,

Abstract: Lipoxygenases, which are widely distributed among plant and animal species, are Fe-containing dioxygenases that act on lipids containing (Z,Z)-pentadiene moieties in the synthesis of compounds with a variety of functions. Utilizing an improved strategy of data collection, low temperature, and synchrotron radiation of short wavelength, the structure of ferrous soybean lipoxygenase L-1, a single chain protein of 839 amino acid residues, has been determined by X-ray crystallography to a resolution of 1.4 A. “…Mammalian lipoxygenases do not have the N-terminal domain present in LOX-1 and other related plant lipoxygenases, thus showing smaller molecular mass (75-80 kDa compared to 94-104 kDa in plants). It has been suggested that the N-terminal domain in LOX-1 makes only a loose contact with the C-terminal domain (9), and that it may be also dispensable for plant lipoxygenases (10), because all the amino acid side chains responsible for catalysis are located in the C-terminal domain.
