Diphyllin Improves High-Fat Diet-Induced Obesity in Mice Through Brown and Beige Adipocytes

Abstract: Brown adipose tissue (BAT) and beige adipose tissue dissipate metabolic energy and mediate nonshivering thermogenesis, thereby boosting energy expenditure. Increasing the browning of BAT and beige adipose tissue is expected to be a promising strategy for combatting obesity. Through phenotype screening of C3H10-T1/2 mesenchymal stem cells, diphyllin was identified as a promising molecule in promoting brown adipocyte differentiation. In vitro studies revealed that diphyllin promoted C3H10-T1/2 cell and primary b… Show more

“…Here, we found that the expression of MFAP5 was significantly down-regulated during adipocyte differentiation and maturation. We verified the sequencing results and demonstrated the function of MFAP5 in adipogenesis by silencing or overexpression in C3H10 and 3T3-L1 cells [29][30][31][32] . As mesenchymal stem cells, C3H10 cells are more primitive than 3T3-L1 cells and have multidirectional differentiation ability.…”

“…The ratio of oleic acid to palmitic acid in the diet can determine the concentration of postprandial thrombosis and fibrinolytic factors in men 47 . There is no direct evidence that cleistanthin B and diphyllin are related to thrombosis, but they have progressive effects in inducing cell apoptosis and improving obesity 48 , 49 . Unfortunately, quercetin, apigenin, luteolin, cynaroside, scopoletin, palmitic acid, cleistanthin B, and diphyllin are not unique compounds in Justicia, but it is undeniable that they contribute to the anti-thrombotic effect of Justicia.…”

“…Duan et al found that diphyllin intervention improved the glucolipid metabolism and insulin sensitivity, as evidenced by the increased thermogenesis in BAT and beige adipose tissues, as well as the enhanced BAT generation and WAT browning. It was speculated that diphyllin acted through inhibiting V-ATPase and intracellular autophagy (Duan et al, 2020). Besides, it is worth mentioning that aryl naphthalene lignans are widely found in various dietary or medicinal plants and have multiple biological activities.…”
