KAP1, a Novel Substrate for PIKK Family Members, Colocalizes with Numerous Damage Response Factors at DNA Lesions

Abstract: The DNA damage response requires a coordinated nucleocytoplasmic cascade of events, which ultimately converge on damaged DNA packaged in chromatin. Few connections between the proteins that remodel chromatin and the proteins that mediate this damage response have been shown. We have investigated the DNA damage-induced phosphorylation of the KRAB-ZFP-associated protein 1 (KAP1), the dedicated corepressor for Krüppel-associated box (KRAB) zinc finger protein (ZFP) proteins. We show that KAP1 is rapidly phosphory… Show more

“…Re-expression of Flag-tagged ATM WT but not Y370F mutant rescued p-CHK2 IRIF ( Figure 4E), indicating that ATM Y370 phosphorylation is also involved in p-CHK2 IRIF formation. Similar results were observed that only restoring expression of Flag-tagged ATM WT but not the Y370F mutant rescued ATM-mediated p-KAP1 [49,50] IRIF in ATM-depleted HeLa cells (Supplementary information, Figure S4G). We further investigated whether ATM Y370 phosphorylation is also involved in S343 phosphorylation of NBS1, as ATM serves as an upstream kinase in the ATM-NBS1-SMC1 signaling for cell cycle checkpoint [51,52].…”

“…This regulation by DNA-PK but not by ATM is striking because many of the proteins targeted during DNA repair, such as H2AX, 53BP1, and KAP1, can be phosphorylated by either ATM or DNA-PK (13,15,34). Although DNA-PK inhibition diminishes PA200 on chromatin by 34-40%, there may also be other regulators (such as ATR) of PA200 responsiveness to IR-induced damage.…”

“…17,18 Phosphorylated KAP1 spreads rapidly throughout chromatin and has been implicated in an ATM kinase-dependent chromatin decondensation within five minutes of exposure to agents that introduce DNA DSBs. Disruption of KAP1 renders cells hypersensitive to agents that introduce DNA DSBs.…”
