Wnt5a regulates distinct signalling pathways by binding to Frizzled2

Abstract: Wnt5a regulates multiple intracellular signalling cascades, but how Wnt5a determines the specificity of these pathways is not well understood. This study examined whether the internalization of Wnt receptors affects the ability of Wnt5a to regulate its signalling pathways. Wnt5a activated Rac in the b-catenin-independent pathway, and Frizzled2 (Fz2) and Ror1 or Ror2 were required for this action. Fz2 was internalized through a clathrin-mediated route in response to Wnt5a, and inhibition of clathrindependent in… Show more

“…Because Wnt5a has been classically shown to inhibit b-catenin signaling via Frizzled-2, we next examined any changes in its expression after PH. 16 A notable increase in Frizzled-2 RNA and protein expression was noted at days 1 to 3 after PH, suggesting an active Wnt5aeFrizzled-2 axis during these times of LR ( Figure 2, B and C).…”

“…Wnt5a bound to Fz2, Fz5 and Fz8 but not to Fz6 and Fz7 (Sato et al, 2010) (data not shown), and Ror2, a single transmembrane protein, is known to function as a Wnt5a receptor (Oishi et al, 2003). To examine which receptor(s) are involved in cell migration of DU145 cells, we depleted Wnt receptors by siRNA (Supplementary Figure S2b).…”
